Job description:

Addition Therapeutics is seeking a highly skilled and motivated Associate Scientist/Scientist to join its HIV Molecular and Cellular Biology team. The successful candidate will play an essential hands-on role in advancing our HIV CAR-T program. The ideal candidate will have experience working in the HIV-1 field preferably in the context of CAR-T cells. The successful candidate will be proficient in multiparameter flow cytometry, including intracellular cytokine staining and analysis. She/he will have experience with lentiviral vector production, transduction of primary T cells, and long-term coculture assays. The individual will work closely with scientists across multiple functional groups to generate, analyze, and integrate in vitro cellular efficacy and molecular readouts that support the advancement of the CAR-T program. She/he will be a highly collaborative, resourceful, and proactive scientist with excellent communication skills and the ability to work effectively in a fast-paced, matrixed environment. This role offers significant learning and growth opportunities in the field of genomic medicines and provides a unique opportunity to explore the multifaceted applications of PRINT™ technology.

Key Responsibilities:

  • Generation of replication competent HIV reporter virus followed by virus titration.
  • Evaluate different cell lines expressing CAR-T target for killing efficiency
  • Infection of primary activated PBMC / T-cells with subsequent flow analysis
  • Establish in vitro HIV challenge assays to quantify CAR-T mediated killing of PBMC.
  • Perform and interpret multiparameter flow cytometry analyses to characterize CAR-T cell expression, target binding, killing, and analyze immune populations.
  • Support molecular efforts such as lentivirus and CAR-T construct cloning, IVTs, etc.
  • Integrate in vitro efficacy, cellular, and molecular readouts to guide program decisions.
  • Troubleshoot experimental challenges and drive improvements in study design and execution.
  • Maintain detailed and organized study records; contribute to study reports and presentations.

Qualifications

  • PhD in Biology, Pharmacology, Immunology, Cancer Biology, or related field, with 4-6 years of relevant research or industry experience..
  • Comfortable working with replication-competent HIV in an appropriate biosafety setting.
  • Experience with culturing primary human PBMCs and isolating T-cell populations
  • Experience assessing in vitro tumor killing/efficacy and CAR-T expansion or persistence.
  • Proficiency in multiparameter flow cytometry and flow cytometry data analysis.
  • Experience with ddPCR, ELISA/MSD, or related molecular/protein assays is preferred.
  • Strong experimental, design, analytical, troubleshooting, organizational, and communication skills.
  • Meticulous attention to detail and strong documentation practices including use of electronic notebook for record keeping.

Education: Bachelor’s or master’s degree in chemistry, Biochemistry or related field with 2-4 years of experience 

Compensation: The salary range for this position is $112,000-$132,000.
